چکیده
Background: Candida albicans is the predominant yeast reported from human infection. Non-albicans species have been recently developed as medically vital fungi. Therefore, it essential to detect and identify pathogens at level prescribe appropriate treatment. Methods: This study assessed two complementary methods, including real-time polymerase chain reaction-high resolution melt (PCR-HRM) reaction-restriction fragment length morphism (PCR-RFLP) with standard PCR Sanger sequencing benchmark. Results: In total, 66 samples were tested, newly-advanced assays more effective displayed comprehensive concordance (66/66, 100%) outcomes. Moreover, accurate economical tests positively advanced by PCR-HRM for C. parapsilosis complexes. Conclusions: Given number of studies performed on comparison sensitivity specificity phenotypic genotypic methods diagnose invasive fungal findings this study, could be stated that correlative PCR-RFLP effectively substitutes conventional reasonable identification. However, supplementary evaluations confirming should carried out a broad range standardize method routine application in medical laboratories.

متن کاملDetection and discrimination of two Brucella species by multiplex real-time PCR and high-resolution melt analysis curve from human blood and comparison of results using RFLP
Objective(s): Rapid and accurate detection of Brucella abortus and Brucella melitensis from clinical samples is so important because antibiotic treatment has major side effects. This study reveals a new method in detection of clinical samples of brucellosis using real-time PCR and high-resolution melt (HRM) curve analysis. Materials and Methods: 160 brucellosis suspicious samples with more tha...
متن کاملA simple and reliable PCR-restriction fragment length polymorphism assay to identify Candida albicans and its closely related Candida dubliniensis
Candida dubliniensis is an emerging pathogen capable of causing superficial as well as systemic infections. Due to its close similarity to C. albcians, conventional methods based on phenotypic traits are not always reliable in identification of C. dubliniensis. In this study, we developed a PCR-restriction fragment length polymorphism (RFLP) assay to identify and discriminate between the two cl...
متن کاملDevelopment of species-specific PCR and PCR-restriction fragment length polymorphism assays for L.infantum/L.donovani discrimination.
Discrimination of Leishmania infantum and L. donovani, the members of the L. (L.) donovani complex, is important for diagnosis and epidemiological studies of visceral leishmaniasis (VL). We have developed two molecular tools including a restriction fragment length polymorphisms of amplified DNA (PCR-RFLP) and a PCR that are capable to discriminate L. donovani from L. infantum. Typing of the com...
متن کاملDirect early identification of Mycobacterium tuberculosis by PCR-restriction fragment length polymorphism analysis from clinical samples.
We attempted to apply the PCR-restriction fragment length polymorphism (PCR-RFLP) technique for the early detection and identification of Mycobacterium tuberculosis directly from clinical samples. PCR-RFLP of hsp65 was applied to the DNA extracted directly from sputum samples (n = 226) collected from 226 patients. We could detect and identify M. tuberculosis in 84.5% of the acid-fast bacillus (...
